Functions & Usage
What is Pemoline used for?
Pemoline is a medicine to treat symptoms attention deficit hyperactivity disorder (ADHD) by stimulating the central nervous system (brain and nerves). It is not yet known how pemoline works.
Pemoline can also be used for purposes other than those listed in the medication guide.
What are the rules for using Pemoline?
Use this medication exactly as directed by your doctor. If you do not understand these directions, ask your pharmacist, nurse, or doctor to explain the instructions for you.
Take each dose with a glass of water.
Pemoline is usually taken once a day in the morning. Follow the doctor's instructions.
In rare cases, pemoline has caused severe liver damage resulting in death or liver transplantation. Tell your doctor right away if you experience nausea, vomiting, stomach pain, unusual fatigue, loss of appetite, yellow skin or eyes, itching, clay-colored stools, or dark colored urine. These symptoms may be early signs of liver damage. Before using pemoline, you will be asked to discuss with your doctor the risks and benefits of this medication and to sign an agreement stating that you understand the risks and benefits. In addition, your doctor will need to monitor your liver function with blood tests before starting therapy with pemoline and every two weeks thereafter.
Follow the rules given by your doctor or pharmacist before starting treatment. If you have any questions, consult your doctor or pharmacist.
How do I store Pemoline?
This medication is best stored at room temperature, away from direct light and damp places. Don't keep it in the bathroom. Don't freeze it. Other brands of this drug may have different storage rules. Observe the storage instructions on the product package or ask your pharmacist. Keep all medicines away from children and pets.
Do not flush medicines down the toilet or down the drain unless instructed to do so. Discard this product when it has expired or when it is no longer needed. Consult your pharmacist or local waste disposal company about how to safely dispose of your product.
Precautions & Warnings
What should be considered before using Pemoline?
In deciding to use this drug, the risks of using the drug must be weighed against its benefits. This is up to you and your doctor. For this drug, the following should be considered:
Allergy
Tell your doctor if you have ever had an unusual or allergic reaction to this medicine or any other medicines. Also tell your health care professional if you have any other types of allergies, such as to food, coloring, preservatives, or animals. For non-prescription products, read the label or ingredients on the package carefully.
Children
Slowing growth rates in children who receive drugs such as pemoline for long periods of time have been reported. Some doctors recommend a drug-free period during treatment with pemoline to help prevent slowed growth.
Pemoline can lead to bad behavior in children with serious mental illnesses.
Is the drug Pemoline safe for pregnant and lactating women?
There is no adequate research on the risks of using this drug in pregnant or breastfeeding women. Always consult your doctor to weigh the potential benefits and risks before using this medication. This drug is included in the risk of pregnancy category B according to the US Food and Drug Administration (FDA). (A = No risk, B = No risk in some studies, C = Possible risk, D = Positive evidence of risk, X = Contraindicated, N = Unknown)
Side effects
What are the possible side effects of Pemoline?
The following are the side effects in descending order of severity in each category associated with Cylert (pemoline):
Heart: There have been reports of liver dysfunction, ranging from asymptomatic reversible elevated liver enzymes to hepatitis, jaundice and dangerous liver failure in patients taking Cylert (see PREVENTION and WARNING).
Hematopoietic: There are isolated reports of aplastic anemia.
Central nerve system: The following CNS effects have been reported with the use of cylert (pemoline): seizures: literature reports suggest that cylert (pemoline) can trigger attacks of Gilles de la Tourette syndrome; hallucinations; movement dyskinesia of the tongue, lips, face and extremities: abnormal oculomotor function including nystagmus and oculogyric crisis; mild depression; dizzy; increased irritability; headache; and sleepy.
Insomnia is the most commonly reported side effect of Cylert (pemoline), usually occurring early in therapy before an optimal therapeutic response. In most of the cases this subsides naturally or responds with a decrease in the dose.
Gastrointestinal: Anorexia and weight loss may occur during the first weeks of therapy. In most of the cases this subsides naturally; weight usually returns within three to six months.
Nausea and stomach pain have also been reported.
Genitourinary: A case of elevated acid phosphatase in association with an enlarged prostate has been reported in a 63 year old man who was treated with Cylert (pemoline) for drowsiness. Acid phosphatase is normalized by discontinuation of Cylert (pemoline) and again increases with reuse.
Others: Growth slowdown has been reported with long-term use of stimulants in children. (See WARNING.) Skin rashes have been reported with Cylert (pemoline).
Mild side effects occurred during the start of treatment with Cylert (pemoline) often returned with continued treatment. If the adverse reaction is significant or protracted, the dose should be reduced or drug use discontinued.
Not everyone experiences the following side effects. There may be some side effects not listed above. If you have concerns about certain side effects, consult your doctor or pharmacist.
Drug Interactions
What medicines can interfere with the drug Pemoline?
Drug interactions can change the performance of your medications or increase the risk of serious side effects. Not all possible drug interactions are listed in this document. Keep a list of all the products you use (including prescription / non-prescription drugs and herbal products) and consult your doctor or pharmacist. Do not start, stop, or change the dose of any medication without your doctor's approval
Although certain drugs should not be used at the same time, in other cases two different drugs can be used together even if interactions are possible. In this case, your doctor may want to change the dose or take other precautions that may be necessary. Tell your health care professional if you are taking any other prescription or nonprescription drugs on the market.
Can certain foods and drinks interfere with the action of the drug Pemoline?
Certain drugs should not be used with meals or when eating certain foods because drug interactions can occur. Consuming alcohol or tobacco with certain drugs can also cause interactions to occur. Discuss your use of drugs with food, alcohol, or tobacco with your health care provider.
What health conditions can interfere with the performance of the drug Pemoline?
The presence of other health problems in your body can affect the use of this drug. Tell your doctor if you have other health problems, such as:
- Drug abuse or dependence (or history) - dependence on pemoline may occur.
- Gilles de la Tourette syndrome or "tics"
- Liver disease
- Mental illness (severe) - Pemoline can make the condition worse
- Kidney disease - higher levels of pemoline in the blood may occur which can increase the chance of side effects

Usual Adult Dose for Attention Deficit Disorder
Pemoline was voluntarily withdrawn from the US market by manufacturers in October 2005 due to a conclusion by the FDA that the overall risk of liver toxicity from pemoline products outweighed the benefits of this drug. The following dosage information applies when this drug is available in the US
Initial dose: 37.5 mg orally every morning.
Maintenance dose: May be increased by 18.75 mg daily at one-week intervals, up to a maximum of 112.5 mg / day. The effective dosage range is 56.25-75 mg for the majority of patients.
What is the dosage of Pemoline for children?
Usual Children's Dose for Attention Deficit Disorder
Pemoline was voluntarily withdrawn from the US market by manufacturers in October 2005 due to a conclusion by the FDA that the overall risk of liver toxicity from pemoline products outweighed the benefits of this drug. The following dosage information applies when this drug is available in the US
> = 6 years:
Initial dose: 37.5 mg orally every morning.
Maintenance dose: May be increased by 18.75 mg daily at one-week intervals, up to a maximum of 112.5 mg / day. The effective dosage range is 56.25-75 mg for the majority of patients.
In what dosages and preparations is Pemoline available?
18.75 mg tablet (white) in bottle 100 (NDC 0074-6025-13);
37.5 mg tablet (orange color) in bottle 100 (NDC 0074-6057-13);
75 mg tablet (tan colored) in bottle 100 (NDC 0074-6073-13).
Cylert (pemoline) chewable tablets are provided as a tonal 37.5 mg monogram (orange colored) tanlet in 100 bottles (NDC 0074-6088-13).
What to do in an emergency or overdose?
In case of an emergency or overdose, contact the local emergency services provider (118/119) or immediately to the nearest hospital emergency department.
What should I do if I forget to take medicine or forget to take medicine?
If you forget a dose of this medicine, take it as soon as possible. However, when it is nearing the time of the next dose, skip the missed dose and return to the usual dosing schedule. Don't double the dose.
